Summary
Allometry describe the relationship between body characteristics and some parameters. In pharmacokinetics allometric scaling could be used
- To predict human behavior from preclinical data before any administration, for example determination of the dose for first in Man study, plasma sample schedule, anticipate elimination and clearance.
- To anticipate and support administration of drug in specific population deriving data from the heathy volunteer’s or clinical data for example pediatric population from adult data, or sub adult population such as Obese or Elderly patient from main cohorts.
Often the main parameter of interest is clearance (a combination of elimination rate and volume of distribution) and allometric extrapolation could be done using various models such as In Vitro/In Vivo Extrapolation Allometric modeling, PBPK models, population approach, etc.… Authorities accept such approaches and provide guideline to help the users.
