Abstract

Ziprasidone is widely used for psychiatric disorders, yet population pharmacokinetic data to guide personalized dosing in Chinese patients were lacking. This retrospective study analyzed 547 concentration measurements from 185 psychiatric patients (with 30 additional patients for external validation) to build a population pharmacokinetic model using Phoenix NLME. Co-administration of lorazepam and valproic acid significantly influenced ziprasidone clearance, and the validated model was used to derive a practical, easy-to-use dosing regimen table. This Chinese-specific population pharmacokinetic model offers a scientific basis for individualizing ziprasidone dosing in patients on common concomitant medications.
