Abstract

Wendan Decoction is a traditional six-herb Chinese medicine formula used to treat hyperlipidemia, though its antioxidant mechanism of action had not been thoroughly characterized through a pharmacokinetic-pharmacodynamic lens. This study developed a PK-PD model in a rat model of high-fat-diet-induced hyperlipidemia, measuring drug component concentrations by UPLC-MS/MS and lipid peroxide levels over time, with modeling performed using Phoenix WinNonlin. Wendan Decoction reduced triglycerides, total cholesterol, and LDL cholesterol in a dose-related manner, with a delayed (counterclockwise hysteresis) relationship between drug concentration and lipid-lowering effect, and components including hesperetin, quercetin, naringenin, and tangeretin appeared to drive the antioxidant activity. The PK-PD model developed here may help guide further investigation of Wendan Decoction’s lipid-lowering mechanism.

Phoenix WinNonlin: PK-PD for Traditional Medicine

Phoenix WinNonlin built the PK-PD model behind this traditional Chinese medicine study, linking herbal component exposure to lipid-lowering effect. As part of a Phoenix platform designed to move teams from data to decision in a fraction of the time, Phoenix WinNonlin’s results can flow directly into PK Submit for CDISC-compliant submission packages.
