Abstract

Fenugreek seed extracts contain bioactive components with reported antidiabetic, antibacterial, and other therapeutic properties, but simultaneous pharmacokinetic characterization of these components had not been established. This study developed and validated an LC-MS/MS method to simultaneously quantify four fenugreek bioactives (4-hydroxyisoleucine, trigonelline, raffinose, and pinitol), then applied it to characterize their oral pharmacokinetic profile in Sprague-Dawley rats using non-compartmental analysis in Phoenix WinNonlin. The validated method enabled rapid, selective quantification of all four analytes from plasma samples. This work provides an analytical and pharmacokinetic foundation for further research into fenugreek-based herbal formulations.

Phoenix WinNonlin: Multi-Analyte PK in One Method

Phoenix WinNonlin’s non-compartmental analysis characterized four fenugreek bioactives simultaneously in this study, built on a newly validated LC-MS/MS method. Within a Phoenix platform that centralizes data, analyses, and templates with built-in collaboration, Phoenix WinNonlin’s output can become submission-ready tables, figures, and listings through TFL Studio.
