Abstract

Preclinical pharmacokinetic assessment of antiseizure medications is essential for understanding their clinical potential, but species differences between mice and rats are not always well characterized. This study combined seizure protection assays with plasma and brain pharmacokinetic analysis (via Phoenix WinNonlin) for several prototype antiseizure drugs, including clobazam, carbamazepine, valproate, and levetiracetam, in both mouse and rat models. Distinct pharmacokinetic profiles were observed between species, including differing clobazam metabolite ratios and drug half-lives, which correlated with differences in seizure protection duration between mice and rats. The study demonstrates how pharmacokinetic analysis can explain species-specific differences in antiseizure drug efficacy and duration of action in rodent models.

Phoenix WinNonlin: Explaining Species Differences

Phoenix WinNonlin’s pharmacokinetic analysis linked species-specific drug half-lives to differences in seizure protection across mouse and rat models in this study. Paired with AI PK Reports, Phoenix WinNonlin can help turn an analysis into a complete PK document in minutes rather than days, within a Phoenix platform used by 100% of the top 20 pharma companies.
