Abstract

Phenylbutazone is a commonly used equine anti-inflammatory drug, but its pharmacokinetics in geriatric horses had not been well characterized despite the potential for age-related changes in drug disposition. This study compared phenylbutazone and its active metabolite oxyphenbutazone following a single intravenous dose in eight young-adult and eight geriatric horses, with pharmacokinetic parameters calculated using Phoenix WinNonlin. No significant differences in phenylbutazone or oxyphenbutazone pharmacokinetic parameters were found between age groups, despite a two-compartment model best describing drug disposition in both. The findings do not support the need for phenylbutazone dose adjustments in clinically healthy geriatric horses compared with younger adults.
