Abstract
Trigonella foenum-graecum (fenugreek) extract contains bioactive compounds effective for treating various conditions, but their pharmacokinetics in diabetic states had not been characterized. This study developed a validated UPLC-MS/MS method to simultaneously quantify trigonelline, 4-hydroxyisoleucine, and diosgenin, then applied it to compare their pharmacokinetics between normal and type 2 diabetic Wistar rats using non-compartmental analysis in Phoenix WinNonlin. The absorption rate of these bioactives increased while their distribution and elimination slowed in diabetic rats compared with normal rats. These findings suggest that diabetic status meaningfully alters the pharmacokinetics of fenugreek’s bioactive components, relevant to their therapeutic use in this population.
