Abstract

Dasatinib, a tyrosine kinase inhibitor used for chronic myeloid leukemia (CML), shows high pharmacokinetic variability, but data characterizing this variability in Chinese patients were scarce. This study used 754 plasma concentrations from 140 Chinese CML patients (with 140 additional samples for external validation) to build a population pharmacokinetic model with Phoenix NLME, examining genetic and physicochemical covariates. Age was significantly associated with dasatinib clearance, with elderly patients showing notably higher exposure in simulation, while no genetic factors were found to be significant covariates. The findings suggest that low-dose dasatinib may be better suited for Chinese patients, with the dose potentially reduced further with increasing age, especially in the elderly.
